PCR is used to detect various Y chromosome loci including the SRY gene located at the testis-determining region on the short arm (Yp11.31); TTTY11 gene located at the proximal region on the short arm (Yp11.2); and DDX3Y gene located at the distal region on the long arm (Yq11.21). Failure to amplify these loci indicates no Y chromosome cell line exists, while if all or part of these sequences amplify, there is evidence for a Y chromosome cell line.
